Underlying Cause Identification

Underlying cause identification is the systematic process of determining why a clinical symptom, sign, or condition occurs rather than merely describing its presentation. Clinicians combine the patient’s history, physical examination, risk factors, and targeted laboratory or imaging findings to develop and test a differential diagnosis, weighing how well each possible cause explains the available evidence. This approach helps distinguish primary disease from contributing conditions, treatment effects, or environmental influences. Identifying the underlying cause supports timely, individualized management, reduces unnecessary interventions, and improves prognosis by directing treatment toward the biological process responsible for the patient’s condition.

This paper describes a protocol that assesses the changes of myofilament Ca2+ sensitivity during contraction in isolated cardiac myocytes from rat heart. Together with cardiac electrophysiology, systolic/diastolic cytosol Ca2+ levels and contraction/relaxation, this measurement is imperative in underpinning the mechanisms mediating cardiac excitation-contraction coupling in healthy and diseased hearts.
