Tol2 Transposase

Tol2 transposase is an enzyme that mobilizes DNA segments from the Tol2 transposable element, providing a powerful tool for introducing genetic material into cells and embryos. It recognizes terminal inverted repeat sequences flanking a cargo sequence, excises that DNA from a donor construct, and integrates it into a new genomic site through a cut-and-paste mechanism. In developmental biology, researchers use Tol2-mediated transgenesis to generate stable reporter lines, express candidate genes, and trace cell lineages in model organisms such as zebrafish. Its efficient genomic integration supports studies of gene regulation, tissue formation, and developmental function, although insertion location can influence transgene expression.
