Geometric Focus Depth

Geometric focus depth is the range of image-plane positions over which an optical system produces an acceptably sharp image according to a specified allowable circle of confusion. It is determined from ray geometry: when the sensor or image surface moves away from the ideal focus plane, rays form a blur circle whose diameter increases with defocus, aperture size, and system geometry. In engineering, this concept guides camera, microscope, machine-vision, and imaging-system design by linking aperture, focal length, working distance, and tolerance requirements to focus accuracy. It helps engineers set alignment tolerances and predict image quality without relying solely on subjective sharpness.

A microchannels-on-a-chip platform was developed by the combination of photolithographic reflowable photoresist technique, soft lithography, and microfluidics. The endothelialized microchannels platform mimics the three-dimensional (3D) geometry of in vivo microvessels, runs under controlled continuous perfusion flow, allows for high-quality and real-time imaging and can be applied for microvascular research.
