Pit Excavation

Pit excavation is the controlled removal of soil and rock to create a below-ground cavity, supporting construction, environmental investigation, and land management. The process involves planning the pit dimensions, removing material in stages, and maintaining stable sidewalls through appropriate excavation methods, shoring, or sloping; groundwater and surface runoff may require additional control. In environmental work, pits provide access for examining soil layers, collecting samples, identifying contamination, and assessing subsurface conditions. Careful excavation limits erosion, sediment movement, habitat disturbance, and worker hazards, making site planning and restoration important components of responsible practice.

Soil Lysimeter Excavation for Coupled Hydrological, Geochemical, and Microbiological Investigations

This study presents an excavation method for investigating subsurface hydrological, geochemical, and microbiological heterogeneity of a soil lysimeter. The lysimeter simulates an artificial hillslope which was initially under homogeneous condition and had been subjected to approximately 5,000 mm of water over eight cycles of irrigation in an 18-month period.

In this video, we describe an in vitro osteoclastic resorption assay on calcium phosphate-coated cell culture plates cultured with human peripheral blood mononuclear cell-derived osteoclasts. The osteoclasts form a characteristic actin ring on the calcium phosphate-coated matrix of the cell culture plate and degrade the matrix within the actin ring region to release calcium and phosphate ions, resulting in the formation of distinct resorption pits within the degraded matrix.

The sampling ring is then placed into the soil to collect the sample. The ring with the sample is then cleaned, labeled, packed, and transferred undisturbed for analysis.
