Nitrocellulose Filter Binding

Nitrocellulose filter binding is a biochemical technique used to detect and measure interactions between nucleic acids and proteins, making it useful for studying gene regulation and molecular recognition. In the assay, a protein or protein complex is incubated with DNA or RNA and passed through a nitrocellulose membrane; the membrane retains the protein and any nucleic acid bound to it, while unbound nucleic acid passes through. Measuring the retained nucleic acid, often with a labeled probe, reveals binding affinity, specificity, and the effects of sequence or experimental conditions. In genetics research, the method helps characterize transcription factors, DNA-binding proteins, RNA-binding proteins, and regulatory interactions.

Quantification of Bacterial Histidine Kinase Autophosphorylation Using a Nitrocellulose Binding Assay

We report and demonstrate an optimized nitrocellulose binding assay that can be used to quantify autophosphorylation of purified bacterial histidine kinases. Our method has several advantages over traditional SDS-PAGE based techniques, providing a valuable alternative for characterizing these important proteins.
