Clinical Grade Preparation

Clinical grade preparation refers to producing biological materials, cells, reagents, or formulations to standards suitable for clinical investigation or patient use, where safety, identity, purity, and consistency are essential. It uses controlled sourcing and processing, documented procedures, contamination control, and release testing to preserve biological activity while meeting predefined quality requirements, often within good manufacturing practice frameworks. In immunology and infection research, this approach supports the preparation of immune-cell products, vaccine components, and assay reagents that can be evaluated reliably in clinical studies. Standardized preparation strengthens reproducibility, traceability, and translation from laboratory findings to diagnostics, prevention, and treatment.

Protocol for propagation of dissociated high grade glioma surgical specimens in serum-free neurosphere medium to select for cells with cancer stem cell phenotype. For specimens that fail to grow as neurospheres, an alternative protocol is suggested. A paraffin embedding technique for maintaining the 3D neurosphere architecture for immunocytochemistry is described.

2025

Here, we present a protocol to evaluate ultrasound-assisted scoliosis-specific exercise in adolescents with mild idiopathic scoliosis, with participants randomly assigned to a training program conducted three times per week for three months, followed by a three-month observation period, providing real-time feedback on intercostal muscle activation and breathing control.

Graded Potential

2025

Graded potentials are localized fluctuations in the cell membrane's electrical charge, commonly found in the dendrites of neurons. The magnitude of these potential changes depends on the strength of the initiating stimulus. In a membrane at its resting potential, a graded potential signifies a voltage shift either above -70 mV or below -70 mV. Graded potentials fall into two categories: depolarizing and hyperpolarizing. Depolarizing graded potentials typically occur when sodium (Na+) or calcium...

Types of Aggregate Grading

2024

Aggregate grading is crucial in economically obtaining a concrete mix with adequate strength, reasonable workability, and minimal segregation. There are four types of aggregate gradation: well-graded, uniformly (or one-sized) graded, gap-graded, and open-graded. Well-graded aggregates include a complete range of necessary size fractions that fit together to create a dense matrix with minimal voids, represented by a smooth, continuous gradation curve. This type of grading ensures good...
