Diet Gel Delivery

Diet gel delivery is a noninvasive method for administering compounds to laboratory animals by incorporating them into a palatable gel that animals consume voluntarily. After the compound is uniformly mixed into the gel, animals receive it ad libitum, while intake, concentration, and body mass can be monitored to estimate exposure and support consistent dosing. In immunology and infection research, this approach can deliver antimicrobials, immunomodulators, or other test substances while reducing handling and gavage-related stress, making it useful for longitudinal studies of host responses, treatment efficacy, and disease progression. Its value depends on verifying compound stability, palatability, and actual consumption.

Convection-enhanced delivery (CED) has been proposed as a treatment option for a wide range of neurological diseases. In order to prepare health care professionals for adoption of CED, accessible training models are needed. We describe the use of agarose gel as such a model of the human brain for testing, research, and training.

Gel Purification

2023

Gel purification is used to recover DNA fragments after electrophoretic separation. DNA recovery from an agarose gel includes three basic steps: binding, washing and eluting from a silica column. DNA is believed to bind to silica in the presence of high salt via a salt bridge. Following binding, DNA is washed of impurities and eluted under low salt conditions disrupting this interaction. This video goes through a step-by-step, generalized procedure for cutting out a band from the gel, gel...

High Fat Diet Feeding and High Throughput Triacylglyceride Assay in Drosophila Melanogaster

This is a high fat diet feeding protocol to induce obesity in Drosophila, a model for understanding fundamental molecular mechanisms implicated in lipotoxicity. It also provides a high throughput triacylglyceride assay for measuring fat accumulation in Drosophila and potentially other (insect) models under various dietary, environmental, genetic or physiological conditions.

DNA Gel Electrophoresis

2023

DNA gel electrophoresis is a technique used for the detection and separation of DNA molecules. An electric field is applied to a gel matrix comprised of agarose, and within the gel, charge particles will migrate and separate based on size. The negatively charged phosphates of the DNA backbone cause DNA fragments to move toward the anode - a positively charged electrode. The video explains the mechanism by which DNA fragments are resolved on an agarose gel, and it provides a step-by-step...
