Enzyme Replacement Therapy

Enzyme replacement therapy (ERT) is a treatment that supplies a functional enzyme to people whose cells lack or produce too little of it, helping prevent harmful substrate accumulation. Typically, a recombinant enzyme is administered intravenously and taken up by cells through receptor-mediated endocytosis, after which it reaches intracellular compartments such as lysosomes and restores deficient catalytic activity. ERT is used primarily for inherited metabolic and lysosomal storage disorders, where it can improve cellular function and limit organ damage. In immunology and infection research, the approach also highlights how enzyme deficiencies disrupt immune-cell metabolism and how anti-drug antibodies can influence treatment efficacy and safety.
