Gasdermin E

Gasdermin E (GSDME) is a pore-forming cell-death protein that links apoptotic signaling to pyroptosis, an inflammatory form of programmed cell death relevant to immunity, infection, and cancer biology. When activated apoptotic caspase-3 cleaves GSDME, its released N-terminal fragment binds acidic phospholipids and oligomerizes in the plasma membrane, forming pores that cause cell swelling, membrane rupture, and release of inflammatory signals. Studying this switch helps explain how infected or stressed cells alert the immune system, how tumors may undergo inflammatory death during treatment, and how GSDME activity could be targeted to influence host defense or limit tissue damage.

This video demonstrates the lactate dehydrogenase assay to detect inflammasome-mediated macrophage death. The macrophages are exposed to lipopolysaccharide for priming, followed by exposure to nigericin to cause inflammasome-induced cell death, which is determined by the lactate dehydrogenase assay.
