Low Density Fraction

Low density fraction refers to the population of cells or particles that remains in the low-density layer or at the interface during density-gradient centrifugation, a separation method based on buoyant density. During centrifugation, components migrate through a density medium until their movement is balanced by the medium’s density, allowing the low-density fraction to be collected separately from denser blood cells and debris. In immunology and infection research, this fraction can enrich circulating immune-cell populations, including low-density granulocytes and other activated or immature myeloid cells, for phenotyping, functional assays, transcriptomic analysis, and investigation of inflammation, host responses, and disease-associated cellular changes.

Electroconvulsive Seizures in Rats and Fractionation of Their Hippocampi to Examine Seizure-induced Changes in Postsynaptic Density Proteins

2017

Electroconvulsive seizure (ECS) is an experimental animal model of electroconvulsive therapy for severe depression. ECS globally stimulates activity in the hippocampus, leading to synaptogenesis and synaptic plasticity. Here, we describe methods for ECS induction in rats and for subcellular fractionation of their hippocampi to examine seizure-induced changes in synaptic proteins.

This fractionation protocol will allow researchers to isolate cytoplasmic, nuclear, mitochondrial, and membrane proteins from mammalian cells. The latter two subcellular fractions are further purified via isopycnic density gradient.

Utilizing Soil Density Fractionation to Separate Distinct Soil Carbon Pools

Soil density fractionation separates soil organic matter into distinct pools with differing stabilization mechanisms, chemistries, and turnover times. Sodium polytungstate solutions with specific densities allow the separation of free particulate organic matter and mineral-associated organic matter, resulting in organic matter fractions suitable for describing the soil response to management and climate change.

Density Gradient Ultracentrifugation

2023

Density gradient ultracentrifugation is a common technique used to isolate and purify biomolecules and cell structures. This technique exploits the fact that, in suspension, particles that are more dense than the solvent will sediment, while those that are less dense will float. A high-speed ultracentrifuge is used to accelerate this process in order to separate biomolecules within a density gradient, which can be established by layering liquids of decreasing density in a centrifuge tube. The...
