Minimal Optimal Peptide Sequence

A minimal optimal peptide sequence is the shortest amino acid sequence that retains a desired biological function, such as immune recognition or antimicrobial activity. Researchers identify it by systematically shortening a candidate peptide or substituting individual residues, then measuring effects on peptide stability, major histocompatibility complex binding, T-cell activation, or pathogen inhibition. In immunology and infection research, this approach helps define antigenic epitopes, identify residues essential for host-pathogen interactions, and distinguish functional motifs from inactive flanking regions. The resulting sequences support vaccine antigen design, diagnostic assays, immune-monitoring tools, and development of targeted peptide-based therapeutics.

This method describes the steps to improve the quality and quantity of sequence data that can be obtained from formalin-fixed paraffin-embedded (FFPE) RNA samples. We describe the methodology to more accurately assess the quality of FFPE-RNA samples, prepare sequencing libraries, and analyze the data from FFPE-RNA samples.

Optimized Minimally Invasive Transscleral Subretinal Injection Technique in Mouse

Source:Tia Rizakos1, Jennifer Geddes-McAlister11Molecular and Cellular Biology Department, University of Guelph. This video demonstrates peptide purification through the STAGE Tip procedure. A miniature column containing resin beads bonded to long hydrocarbon chains facilitates hydrophobic interaction-based purification. Upon loading a pre-digested peptide sample into the column, the peptide fragments bind to the resin beads, allowing impurities to pass through. An elution buffer that disrupts...

Immunoaffinity Based Extraction of Ubiquitinylated Peptides: A Technique to Selectively Extract Ubiquitin Tagged Peptides from Purified Peptide Fractions

The comparison and optimization of two plant organellar DNA enrichment methods are presented: traditional differential centrifugation and fractionation of the total gDNA based on methylation status. We assess the resulting DNA quantity and quality, demonstrate performance in short-read next-generation sequencing, and discuss the potential for use in long-read single-molecule sequencing.
