Nasopharyngeal Dissemination

Nasopharyngeal dissemination is the spread of a microorganism within or from the nasopharynx, the upper-airway region behind the nose, and it matters because this site can support colonization, transmission, and progression of infection. Pathogens may adhere to respiratory epithelial cells, persist in mucus, overcome mucociliary clearance and local immune defenses, and reach adjacent airway tissues or new hosts through respiratory secretions. In immunology and infection research, analyzing these steps clarifies how host barriers and immune responses limit or enable spread. This knowledge supports evaluation of transmission risk, disease progression, vaccines, and interventions that reduce respiratory pathogen dissemination.

Methods to examine contributions of the nasopharyngeal-associated lymphoreticular tissues (NALT) to nasal and systemic immune responses of mice to intranasal vaccines are described. We demonstrate a surgical procedure to establish a NALT-dependent mouse model and ex vivo cultures of extracted NALT.
