P38 Mapk Pathway

The p38 MAPK pathway is a stress-responsive intracellular signaling cascade that regulates inflammation, immunity, cell survival, and responses to environmental or cellular challenges. In response to microbial products, cytokines, or cellular stress, upstream kinases activate MKK3 and MKK6, which phosphorylate p38 MAPK and enable it to regulate downstream kinases and transcription factors. In immunology and infection research, this pathway helps explain how macrophages and other immune cells produce inflammatory mediators and respond to pathogens. Mapping p38 signaling supports investigations of host-pathogen interactions, inflammatory disease mechanisms, and potential therapeutic strategies for modulating excessive immune responses.

At the subcellular level, signaling events are dynamically modulated by developmental and environmental cues. Here we describe a protocol that employs the tobacco transient expression system to monitor dynamic protein-protein interaction and to disclose spatial organization of signal transduction in plant cells.

MAPK Signaling Cascades

2023

Mitogen-activated protein kinase, or MAPK pathway, activates three sequential kinases to regulate cellular responses such as proliferation, differentiation, survival, and apoptosis. The canonical MAPK pathway starts with a mitogen or growth factor binding to an RTK. The activated RTKs stimulate Ras, which recruits Raf or MAP3 Kinase (MAPKKK), the first kinase of the MAPK signaling cascade. Raf further phosphorylates and activates MEK or MAP2 Kinases (MAPKK), which in turn phosphorylates MAP...

Temporal Quantification of MAPK Induced Expression in Single Yeast Cells

Two complementary methods based on flow cytometry and microscopy are presented which enable the quantification, at the single cell level, of the dynamics of gene expression induced by the activation of a MAPK pathway in yeast.

C4 Pathway and CAM

Most plants use the C3 pathway for carbon fixation. However, some plants, such as sugar cane, corn, and cacti that grow in hot conditions, use alternative pathways to fix carbon and conserve energy loss due to photorespiration. Photorespiration is the process that occurs when the oxygen concentration is high. Under such conditions, the rubisco enzyme in the Calvin cycle binds O2 instead of CO2, which halts photosynthesis and consumes energy. C4 Pathway The C4 pathway is used by plants such as...

Extracting Host Cell Lysate from Infected Host Cells Cultured in a Permeable Membrane System

2025

This video demonstrates a permeable membrane insert system to investigate the impact of bacterial toxins on host cells in a controlled and isolated manner. This approach allows for the selective exposure of the cells to the toxins while maintaining a physical separation that mimics the natural host-pathogen interaction.
