Rabies Virus Detection

Rabies virus detection comprises laboratory methods used to identify rabies virus or the immune response it triggers, supporting rapid diagnosis of a nearly always fatal infection after symptom onset. Testing may detect viral antigen in nervous tissue by direct fluorescent antibody assays, amplify viral RNA with reverse-transcription polymerase chain reaction, or measure rabies-neutralizing antibodies in serum or cerebrospinal fluid. In immunology and infection research, these approaches help distinguish active infection from prior vaccination, guide post-exposure management, and monitor immune responses. Reliable detection also supports surveillance, outbreak investigation, vaccine evaluation, and studies of viral spread through the nervous system.

An Immunohistochemistry Staining for the Detection of Rabies Virus Antigens

2025

This video demonstrates immunohistochemistry staining for rabies virus antigen detection in infected mouse brain tissue. The process includes deparaffinization and alcohol treatment, followed by proteolytic treatment to enhance antigen accessibility. Immunohistochemistry staining reveals red precipitates, confirming the presence of rabies antigens in the tissue section.

Source: Zaeck, L., et al. High-Resolution 3D Imaging of Rabies Virus Infection in Solvent-Cleared Brain Tissue. J. Vis. Exp. (2019). This video demonstrates the process of preparing and imaging mouse brain tissue infected with the rabies virus (RABV) to evaluate infection progression. It outlines key steps, including tissue staining with fluorescent markers, dehydration, clearing, and mounting for confocal laser scanning microscopy, culminating in the generation of a three-dimensional...
