Sted Microscopy

STED microscopy, or stimulated emission depletion microscopy, is a super-resolution imaging technique that reveals cellular structures beyond the diffraction limit of conventional light microscopy. It uses a focused excitation beam together with a doughnut-shaped depletion beam to suppress fluorescence around the focal spot, restricting signal emission to a nanoscale region; scanning this spot across a labeled specimen produces a high-resolution image. In immunology and infection research, STED microscopy can visualize receptor organization, immune-cell contacts, pathogen entry, and host–pathogen interactions in fixed or living samples. These observations clarify spatial mechanisms that influence immune signaling, microbial invasion, and disease progression.

We present a protocol for using STED microscopy to simultaneously image actin structures, microtubules, and microtubule plus-end binding proteins in B cells that have spread on coverslips coated with antibodies to the B-cell receptor, a model for the initial phase of immune synapse formation.

Here we illustrate the protocol for imaging by two-color STED nanoscopy the cytotoxic immune synapse of NK cells recapitulated on glass. Using this method we obtain sub-100 nm resolution of synapse proteins and the cytoskeleton.

Correlative Light- and Electron Microscopy Using Quantum Dot Nanoparticles

A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of epoxy embedded human pathology tissue. We employ commercial antibody fragment conjugated QDs that are visualized by widefield fluorescence light microscopy and transmission electron microscopy.
