Tcid50 Measurement

TCID50 measurement is a virological assay that estimates the quantity of infectious virus required to produce infection in 50% of inoculated tissue-culture wells. In practice, researchers prepare serial virus dilutions, expose replicate cell cultures, incubate them under controlled conditions, and score infection through cytopathic effects or other assay-defined signals; statistical methods then identify the dilution corresponding to the 50% endpoint. This measurement provides an infectious-virus titer rather than a count of total particles, supporting comparisons among viral stocks, evaluation of sample infectivity, and standardization of immunology and infection experiments, including virus neutralization studies and pathogen characterization.

Optimized sampling protocols and the development of new wipe materials can be facilitated by standardized measurements of collection efficiency from wipe-sampling. Our approach for sampling trace explosives uses an automated device to control speed, force, and distance during wipe-sampling followed by extraction of collected explosives.
