Western Immunoblotting

Western immunoblotting is a laboratory method used to detect and characterize specific proteins in complex biological samples, making it valuable for studying immune responses and infectious disease. Proteins are separated by size through gel electrophoresis, transferred to a membrane, and recognized by primary antibodies; a labeled secondary antibody then produces a measurable signal that indicates the target protein’s presence and relative abundance. In immunology and infection research, the technique helps identify pathogen antigens, assess host signaling proteins, compare protein expression between conditions, and confirm results from other assays. Protein size and antibody specificity also support interpretation of molecular changes during infection or immune activation.

The Western Blot

Western Blotting is used to identify the presence of specific proteins in electrophoretically separated samples. Following separation by a technique known as sodium dodecyl sulfate polyacrylamide gel electrophoresis, or SDS-PAGE, western transfer is used to move proteins from a polyacrylamide gel onto a piece of membrane which traps the proteins in their respective locations. Next, the membranes are probed with antibodies in a process called immunboblotting. Immunoblotting uses...
