Zbp1-dependent Necroptosis

ZBP1-dependent necroptosis is an inflammatory form of programmed cell death in which the nucleic acid sensor Z-DNA binding protein 1 (ZBP1) detects abnormal endogenous or infection-associated nucleic acids and triggers cell destruction. After activation, ZBP1 uses homotypic RHIM interactions to recruit and activate receptor-interacting protein kinase 3 (RIPK3), which phosphorylates mixed lineage kinase domain-like protein (MLKL); MLKL then disrupts the plasma membrane, causing cell rupture and release of inflammatory signals. In immunology and infection, this pathway helps eliminate pathogen-infected cells, particularly during viral infection, while its dysregulation can intensify tissue inflammation and influence host-pathogen interactions.
