Embryo Mortality

Embryo mortality is the loss of an embryo before it can continue normal development, an outcome important in reproductive medicine because it affects fertility, miscarriage risk, and pregnancy success. It can result when errors in chromosome number or cell division impair embryonic development, or when implantation and early placental support fail to sustain the embryo. Studying embryo mortality helps clinicians and researchers evaluate early pregnancy loss, assess embryo quality during assisted reproductive technologies, and investigate maternal, genetic, and environmental contributors. This knowledge supports improved counseling, laboratory culture and selection practices, and strategies for addressing recurrent reproductive failure.

Here we present a protocol for assessing seed survivorship, germination and dormancy under field conditions using buried, labeled seed strips and tetrazolium chloride (TZ) viability testing.

Assessing Pathogenic Bacteria–Induced Mortality in C. elegans Larvae

Source: Anderson, Q. L., et al. A High-throughput, High-content, Liquid-based C. elegans Pathosystem. J. Vis. Exp. (2018)The video demonstrates a liquid-based assay where genetically modified C. elegans larvae are exposed to pathogenic bacteria in a microplate. Bacterial toxins secreted into the medium induce host stress and death. A fluorescent dye stains dead worms, allowing imaging-based quantification of mortality to assess genetic susceptibility or resistance.

Mosquito Feeding Assay: A Method to Evaluate Test Compound Effects on Mortality and Fecundity of Adult Female Mosquitoes

Microinjection of Live Drosophila Embryos: Early Delivery of Reagents to the Developing Embryo

2023

This video describes how to perform microinjections that deliver reagents into live Drosophila embryos. Researchers can use this method to introduce exogenous compounds like nucleic acids and proteins during the early stages of embryo development. The featured protocol demonstrates how to set up and perform the procedure for injection of any soluble reagent–here, a fluorescent protein for live imaging experiments.
