In Vitro Cytotoxicity

In vitro cytotoxicity is the evaluation of whether a substance damages or kills cells maintained outside the body, providing an early measure of biological safety and activity. In controlled cell cultures, researchers expose cells to defined concentrations of a drug, chemical, biomaterial, or other test agent, then assess outcomes such as cell viability, membrane integrity, metabolic activity, or cell death. In medicine, these studies support drug discovery, toxicity screening, evaluation of medical devices and biomaterials, and selection of doses for further testing. Results can reveal concentration-dependent effects and help prioritize candidates before animal studies or clinical research.

The protocol reported here allows the evaluation of the efficacy of photodynamic therapy (PDT) in cell lines and the optimization of the PDT settings before applying the therapy in animal models.

An In Vitro Assay to Assess CAR T Cell Cytotoxicity against Tumor Spheroids

2025

This video demonstrates an in vitro assay evaluating the cytotoxic effects of CAR T cells on 3D tumor spheroids expressing fluorescent proteins. This assessment utilizes the Annexin V red fluorescent probe, which binds to exposed phosphatidylserine residues on the outer leaflet of apoptotic cancer cells after interactions with CAR T cells. The red fluorescence emitted by the probe aids in distinguishing these cells from healthy ones emitting green fluorescence.

An In Vitro Assay for Evaluating Natural Killer Cell Cytotoxicity against Cancer Cells

2025

This video demonstrates performing a lactate dehydrogenase release assay to determine the cytotoxicity of natural killer (NK) cells against the target cancer cells. The assay quantifies the release of the cytoplasmic lactate dehydrogenase enzyme from cancer cells by measuring the absorbance of the red-colored product formed.

An In Vitro Assay to Study the Cytotoxic Capability of Cytokine-Induced Killer Cells

2025

This video demonstrates an in vitro technique to measure the cytotoxic ability of cytokine-induced killer (CIK) cells against cancer cells. Upon co-culturing target cancer cells with CIK cells, the dead cancer cells are identified using a fluorescent viability dye and are quantified using flow cytometry, estimating the cytotoxic effect of CIK cells.

An In Vitro Assay for Studying the Cytotoxicity of Pre-Activated CD8+ T Cells against Cancer Cells

2025

This video showcases an apoptosis assay in cancer cells induced by pre-activated effector T cells. By co-culturing these cells with cancer cells marked with fluorescent nuclei, a caspase-cleavable fluorogenic substrate is added. Apoptotic cancer cells will display a different color in their nuclei, leading to dual fluorescence. This dual fluorescence is indicative of cancer cells undergoing apoptosis.
