Luciferase Complementation

Luciferase complementation is a bioluminescence-based technique that detects molecular interactions by linking complementary fragments of a luciferase enzyme to proteins of interest. When those proteins interact, the luciferase fragments are brought together and reconstitute an active enzyme, which produces measurable light after reacting with its substrate. In medicine, this method helps monitor protein-protein interactions, receptor signaling, and intracellular pathway activity in living cells. Its sensitivity and compatibility with high-throughput formats also support drug discovery by identifying compounds that disrupt or enhance disease-relevant interactions, providing quantitative evidence for therapeutic mechanism and response.

This video demonstrates the split luciferase complementation assay to detect protein-protein interactions. In this assay, the proteins of interest are tagged to small and large fragments of the luciferase enzyme. When the proteins interact, the large and small fragments combine to form an active enzyme complex, which in the presence of a specific substrate, releases bright luminescence that can be measured.

A Split-Luciferase Complementation Assay for Temporally Resolved Measurement of Tau Clearance in Microglia

We describe a sensitive, time-resolved quantitative platform to measure clearance of extracellular tau in human induced pluripotent stem cell (iPSC)-derived microglia using a microplate-based luminescence system. This approach enables sensitive detection of tau degradation kinetics in a microwell format, allowing for high-throughput interrogation of tau handling under various experimental conditions.
