Murine Islet Culture

Murine islet culture is the laboratory isolation and maintenance of pancreatic islets from mice, providing a controlled model for studying endocrine function and diabetes. Researchers typically digest pancreatic tissue with collagenase, separate islets from exocrine material, and sustain them in nutrient-defined culture medium under controlled temperature and carbon dioxide conditions. Cultured islets enable assessment of glucose-stimulated insulin secretion, beta-cell viability, and responses to hormones, inflammatory signals, or candidate therapies. Because mouse islets retain coordinated interactions among beta, alpha, and other endocrine cells, this system supports investigations of diabetes pathophysiology, drug efficacy, and mechanisms regulating blood-glucose homeostasis.

In this video, we describe a procedure to infuse pancreatic islet cells into the hepatic portal vein, followed by application of gel foam to successfully prevent bleeding. Postoperative bleeding is a common cause of high mortality during islet transplantation.

Pancreatic islets, also called the Islets of Langerhans, are a cluster of endocrine cells which produces hormones for glucose regulation and other important biological functions. This protocol explains a method for isolating pancreatic islets by first perfusing collagenase via Ampulla of Vater to digest the exocrine pancreatic tissue and a simplified density gradient centrifugation to purify the islets from mice.

We describe a protocol for visualization of insulin exocytosis in intact islets using pHluorin, a pH-sensitive green fluorescent protein. Isolated islets are infected with adenovirus encoding pHluorin coupled to the vesicle cargo neuropeptide Y. This allows for the detection of insulin granule fusion events by confocal microscopy.
