Ringer's Lactate Solution

Ringer's Lactate Solution is an isotonic balanced crystalloid used to restore extracellular fluid volume and replace electrolytes in clinical medicine. It contains sodium, chloride, potassium, calcium, and lactate in concentrations designed to approximate plasma, while the lactate component is metabolized primarily by the liver to generate bicarbonate and support acid–base balance. Administered intravenously, it can improve circulating volume during fluid loss associated with surgery, trauma, burns, or dehydration. Its electrolyte composition makes it useful for resuscitation and maintenance, although clinicians select it according to the patient’s renal, hepatic, cardiac, and metabolic status and the compatibility of concurrent therapies.

This video describes a colorimetric assay for lactate measurement in cell lysates by using the lactate dehydrogenase enzyme. This enzyme converts lactate into pyruvate, along with the conversion of NAD to NADH that reacts with tetrazolium dyes and gives a colored product that can be measured spectrophotometrically to quantitate the lactate.
