Sla Mismatched Yucatan Mini Pigs

SLA-mismatched Yucatan miniature pigs are porcine research models in which donor and recipient animals differ in swine leukocyte antigen (SLA) genes, the pig equivalent of the major histocompatibility complex. When tissues or organs move between animals with different SLA haplotypes, recipient T cells recognize donor antigen-presenting molecules as foreign, activating cellular and antibody-mediated immune responses that may damage the graft. These models support preclinical transplantation research by enabling evaluation of graft rejection, immunosuppressive strategies, and engineered tissues or organs. Their defined genetic background and clinically relevant immune responses can strengthen the assessment of transplant approaches before human studies.

Surgical Angiogenesis in Porcine Tibial Allotransplantation: A New Large Animal Bone Vascularized Composite Allotransplantation Model

2017

Currently any kind of vascularized composite allotransplantation depends on long-term-immunosuppression, difficult to support for non-life-critical indications. We present a new porcine tibial VCA model that can be used to study bone VCA and demonstrate the use of surgical angiogenesis to maintain bone viability without the need of long-term immune-modulation.

Organisms are capable of detecting and fixing nucleotide mismatches that occur during DNA replication. This sophisticated process requires identifying the new strand and replacing the erroneous bases with correct nucleotides. Mismatch repair is coordinated by many proteins in both prokaryotes and eukaryotes. The Mutator Protein Family Plays a Key Role in DNA Mismatch RepairThe human genome has more than 3 billion base pairs of DNA per cell. All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Acclimation and Positive Reinforcement Training Pigs are social animals that respond best to calm, low-stress handling techniques. Acclimation and positive reinforcement training are important refinement strategies recommended...
