Drosophila Egg-laying

Drosophila egg-laying is the regulated behavioral process by which female fruit flies select suitable sites and deposit eggs, providing a tractable model for studying how animals make reproductive decisions. The behavior integrates external chemosensory and mechanosensory cues with internal reproductive state through neural circuits connecting the brain, ventral nerve cord, and egg-laying apparatus. Researchers use changes in oviposition choice, timing, and egg deposition to investigate sensory integration, motivation, neuromodulation, and behavioral plasticity. Because these circuits are accessible to genetic, imaging, and behavioral methods, Drosophila egg-laying helps reveal general principles of neural control that may apply across animal species.

This protocol describes a high throughput assay for testing egg-laying preferences of Drosophila melanogaster at single-animal resolution. This assay provides a simple, efficient, and scalable platform to identify genes and circuit components that control a simple decision-making process.

Drosophila Egg-Laying Preference Assay: A Method to Test Decision-Making in Flies

2023

Drosophila females choose between substrates and select sites that are attractive for egg-laying. This video describes a behavioral assay that tests their decision-making, called the egg-laying preference assay. The featured clip demonstrates the procedure with a custom-built setup, which enables researchers not only to test the behavior of many females at the same time but is also compatible with video recording and optogenetic setups.

Drosophila Egg Collection and Dechorionation: A Method to Remove the Outermost Egg Layer

2023

The Drosophila embryo is surrounded by protective outer membranes. To gain access to the embryo, the chorion–the outermost of the membranes–is often removed. This video describes a commonly used method of dechorionation using sodium hypochlorite. The example protocol demonstrates the procedure while employing sterile techniques necessary for axenic or gnotobiotic animal rearing.
