Drosophila Larval Nmj

The Drosophila larval neuromuscular junction (NMJ) is a glutamatergic synapse where motor neurons communicate with skeletal muscle, providing a well-characterized model for studying neural signaling. When an action potential reaches the presynaptic terminal, calcium influx triggers synaptic vesicle fusion and glutamate release, which activates receptors on the muscle and produces a postsynaptic response. Its accessible anatomy, defined circuit, and powerful genetic tools allow researchers to examine synaptic development, transmission, plasticity, and homeostatic regulation. Studies of the larval NMJ also clarify mechanisms relevant to neurological disease and help connect molecular changes at synapses with effects on motor function.

Here we describe electrophysiological methods for measuring synaptic transmission at the neuromuscular junction of Drosophila larva. Evoked release is initiated artificially by stimulating the motor neuron axons, and transmission through the NMJ can be measured by the postsynaptic response evoked in the muscle.

Drosophila Larval IHC

Immunohistochemistry (IHC) is a technique used to visualize the presence and location of proteins within tissues. Drosophila larvae are particularly amenable to IHC because of the ease with which they can be processed for staining. Additionally, the larvae are transparent, meaning that some tissues can be visualized without the need for dissection. In IHC, proteins are ultimately detected with antibodies that specifically bind to “epitopes” within the protein of interest. In order to preserve...
