Fda Pi Double Staining

FDA-PI double staining is a fluorescence-based cell viability assay that distinguishes living and membrane-compromised cells, making it useful for evaluating neuronal health in neuroscience research. Fluorescein diacetate (FDA) enters cells and is hydrolyzed by intracellular esterases to produce green fluorescence in viable cells, whereas propidium iodide (PI) penetrates cells with damaged plasma membranes and binds nucleic acids, producing red fluorescence. Researchers use fluorescence microscopy or image-based quantification to assess neurotoxicity, injury, apoptosis-associated membrane damage, and the effects of drugs or experimental treatments. The method provides a rapid visual measure of cell survival in cultures and tissue-derived neural models.
