Flight Initiation Assay

The Flight Initiation Assay is a behavioral method that measures how an animal begins an escape response after detecting a threatening or aversive stimulus. Researchers present a controlled stimulus and quantify responses such as flight-initiation latency, movement speed, trajectory, and escape distance, linking sensory detection to defensive motor output. In neuroscience, this assay helps characterize neural circuits that regulate threat perception, arousal, decision-making, and locomotion. Comparing performance across experimental conditions, genetic backgrounds, or pharmacological treatments can reveal how brain systems control defensive behavior and identify changes associated with neurological dysfunction.

A Simple Flight Mill for the Study of Tethered Flight in Insects

Flight in insects is influenced by a number of factors and the propensity to disperse is an important variable in understanding insect ecology and biological control strategies. We describe the construction and use of a simple, relatively inexpensive, and flexible flight mill for measuring parameters of tethered flight in insects.

This video illustrates the initiation of polymicrobial biofilm formation on virus-infected cells by bacteria and fungi. Herpes simplex virus infection impedes bacterial attachment to the cells, preventing bacterial-fungal co-localization. In contrast, uninfected cells display bacterial-fungal co-localization and initiation of biofilm formation.

This protocol uses three-dimensional (3D) printers and laser cutters found in makerspaces in order to create a more flexible flight mill design. By using this technology, researchers can reduce costs, enhance design flexibility, and generate reproducible work when constructing their flight mills for tethered insect flight studies.
