Incubator Culture Conditions

Incubator culture conditions are the controlled environmental parameters used to maintain living cells or tissues outside the organism, making them essential for reproducible neuroscience experiments. Temperature, humidity, gas exchange, and carbon dioxide concentration are regulated to support metabolism and preserve culture-medium pH through the bicarbonate buffering system, while sterile handling limits contamination. In neuronal cultures, these conditions help sustain cell viability, differentiation, synaptic development, and network activity for studies of neurodevelopment, disease mechanisms, and drug responses. Consistent monitoring and adjustment of incubator settings improve experimental reliability and enable meaningful comparisons across cultures and laboratories.

We describe an extended fear-conditioning protocol that produces overtraining and fear incubation in rats. This protocol entails a single training session with 25 tone-shock pairings (i.e., overtraining) and a comparison of conditioned freezing responses during context and cue tests 48 h (short-term) and 6 weeks (long-term) after training.
