Absolute Pressure

Absolute pressure is the total pressure exerted by a fluid or gas, measured relative to a perfect vacuum, making it an unambiguous pressure scale used in physics. It combines atmospheric pressure with gauge pressure: P_abs = P_gauge + P_atm, while pressure sensors and manometers determine values by comparing a system with a reference pressure. Because atmospheric pressure changes with altitude and weather, distinguishing absolute from gauge pressure is essential when applying gas laws, analyzing thermodynamic processes, and predicting boiling or cavitation conditions. This concept supports accurate measurement and safe design of engines, vacuum equipment, pipelines, and laboratory experiments.

The laser heated diamond anvil cell combined with synchrotron micro-diffraction techniques allows researchers to explore the nature and properties of new phases of matter at extreme pressure and temperature (PT) conditions. Heterogeneous samples can be characterized in situ under high pressure by 2D mapping and combined powder, single-crystal and multigrain diffraction approaches.

This protocol describes a method to simultaneously measure cytosolic, free calcium [Ca2+]i and vessel diameter in contracting lymph vessels in real time and then calculate absolute Ca2+ concentrations as well as contractility/rhythmicity parameters.
