The DNA Helix

- [Anlatan] Dioksiribonükleik asit veya DNA, 2 polinükleotid zincirden meydana gelir.

<h4>Detaylı DNA Yapısı</h4>

Her DNA dizisi, şeker deoksiribozu, bir fosfat grubu ve dört azot içeren bazdan birini içeren nükleotitler adı verilen alt birimlerden oluşur: adenin (A), guanin (G), sitozin (C) ve timin (T). Adenin ve guanin, hepsi iki halkalı yapılar içeren pürinler adı verilen daha büyük bir kimyasal sınıfın üyeleridir. Sitozin ve timin, pirimidin adı verilen tek halkalı yapılar grubuna aittir.

Aynı iplikçikteki bitişik nükleotitler, fosfodiester bağları ile kovalent olarak bağlanır. DNA molekülünün iki ipliği, çift sarmal adı verilen yay benzeri bir yapıya sıkıca sarılır. Bununla birlikte, çift sarmal mükemmel simetrik değildir. Bunun yerine, yapıda düzenli olarak oluşan oluklar vardır. Ana oluk, şeker-fosfat omurgalarının nispeten uzak olduğu yerlerde oluşur. <h4>DNA Yapısının Keşfi: Kısa Bir Tarihçe</h4>

DNA yapısının keşfinin hikayesi, İsviçreli bilim adamı Friedrich Miescher'ın "nüklein" olarak adlandırdığı bir maddeyi keşfettiği 1869'da başlıyor. Beyaz kan hücrelerinden protein çıkarma sürecinde, Miescher nispeten yüksek fosfor içeriğine sahip beklenmedik bir madde buldu. Ne olduğunu bilmiyordu, ama biyolojik olarak önemli olabileceğinden şüpheleniyordu. Miescher haklıydı, ancak bilimsel topluluğun içgörülerini tam olarak takdir etmesi onlarca yıl sürdü.

Bir sonraki kritik keşif Rus biyokimyacı Phoebus Levene tarafından yapıldı. 1919'da Levene, nükleik asit olarak bilinen nükleinin, polinükleotid olarak adlandırdığı molekül zincirlerinden oluştuğunu öne sürdü. Levene'nin önerisi, bireysel nükleotidlerin bir fosfat grubu, bir şeker ve azot içeren bir bazdan oluştuğunu bulduğu Maya üzerine yaptığı araştırmalardan kaynaklandı. Avusturyalı biyokimyacı Erwin Chargaff, Levene'nin çalışmalarını genişletti. 1940'ların sonlarında çalışan Chargaff önemli bir bulgu yaptı: DNA'daki adenin miktarı her zaman kabaca timin miktarına eşittir ve guanin miktarı her zaman kabaca sitozin miktarına eşittir. 1950'lerin başında, Amerikalı biyolog James Watson ve İngiliz fizikçi Francis Crick, DNA'nın üç boyutlu yapısını keşfetmek için baş rakibi Amerikan Linus Pauling ile yarışıyorlardı. Chargaff'ın çalışmalarına dayanarak, DNA'nın fiziksel modellerini oluşturmak için fizik, matematik ve kimya bilgisini kullandılar.
