Membrane Lipids

Zarda üç tip lipit bulunur – fosfogliseritler, sfingolipidler ve steroller.
Gliserofosfolipidler olarak da bilinen fosfogliseritler, bir hidrofilik fosfat grubuna ve genellikle 16 ila 22 karbon atomu uzunluğunda iki hidrofobik yağlı asil zincirine sahip bir gliserol omurgası içerir.
Hücre zarında bulunan en yaygın fosfogliseritlerden bazıları, kolin, etanolamin, serin ve inositolün fosfat grubuna bağlanmasıyla oluşan fosfatidilkolin, fosfatidiletanolamin, fosfatidilserin ve fosfatidilinositoldür.

Kalan hidroksil seramid grubu, sırasıyla sfingomyelin veya glikolipidler oluşturan fosfokolin veya karbonhidratlar gibi diğer kimyasal gruplarla esterler oluşturabilir.
Üçüncü tip zar lipitleri, kolesterol gibi steroller bağımsız olarak bir lipit çift tabakası oluşturamaz. Bunun yerine fosfolipid çift tabakaları içinde iç içe geçerler ve zar için bir akışkanlık tamponu görevi görürler.

Lipitler tüm biyolojik membranların önemli bir bileşenidir. Fizyolojik pH'da fosfatidilserin negatif yüklüyken diğer üçü nötr kalır. Membran lipitlerinde, yağ asitlerinde bulunan karbon atomlarının sayısı ve doygunluk dereceleri farklılık gösterebilir. Yağ asitleri çift bağ içermeyen tamamen doymuş, yalnızca bir çift bağ içeren tekli doymamış veya birden fazla çift bağ içeren çoklu doymamış olabilir. Glikolipitler, her zaman membranın dış veya hücre dışı tarafında bulunan, fizyolojik açıdan önemli bir membran lipitleri sınıfını içerir. Ökaryotlarda hem plazma membranında hem de hücre içi membranlarında bulunurlar. Basit bir glikolipit olan galaktoserebrosid, sfingomiyelin ile birlikte miyelin kılıflarında bol miktarda bulunur. En karmaşık glikolipitler olan gangliosidler, sialik asit varlığı nedeniyle negatif yüklüdür ve sinir hücresi membranlarındaki toplam lipitlerin %10'unu oluşturabilir. Gangliosidler viral ve genetik hastalıklardaki rolleri açısından incelenmektedir. Gangliosid GM1, Kolera toksini için hücre yüzeyi reseptörü görevi görürken, genetik bir hastalık olan Tay-Sachs hastalığında GM2 gangliosidlerinin aşırı birikimi gözlenir. Membran lipitlerinin bir başka sınıfı da sterollerdir. Bitkiye özgü steroller, bitki plazma membranın önemli bir bileşenini oluşturur ve toplam membran lipitlerinin %30-50'sini oluşturabilir. Ökaryotik membranlarda toplam lipitlerin neredeyse %50'sini oluşturan kolesteroller de mevcuttur. İnsan hücre membranlarındaki kolesterolün yapısı, mantar membranlarının ortak bir lipit bileşeni olan ergosterolden biraz farklıdır.

Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
