Distance Restraints

Distance restraints are limits placed on the separation between selected atoms or molecular groups, helping researchers determine and evaluate three-dimensional biomolecular structures. In biochemistry, they are often derived from nuclear magnetic resonance measurements, especially nuclear Overhauser effects, whose signal intensities depend strongly on the distance between nuclei; these data are converted into approximate upper or lower distance bounds for structure calculation and refinement. Distance restraints guide computational models toward conformations consistent with experimental observations, supporting studies of protein folding, molecular flexibility, ligand binding, and protein interactions. They also provide a framework for comparing structural models with solution-phase behavior.
