Inorganic Carbon Sequestration

Inorganic carbon sequestration is the capture and long-term storage of carbon dioxide or dissolved inorganic carbon in stable chemical forms, helping limit its return to the atmosphere. In biochemistry and geochemistry, carbon dioxide equilibrates with water to form carbonic acid, bicarbonate, and carbonate; these species can react with calcium or magnesium ions and precipitate as solid carbonate minerals, or remain stored in alkaline aqueous environments. This chemistry underpins mineral carbonation, engineered carbon-removal systems, and studies of ocean carbon cycling. Understanding reaction rates, pH, alkalinity, and mineral stability helps researchers evaluate storage capacity, permanence, and environmental effects.

2025

The video demonstrates an assay for assessing bulk autophagic sequestration activity in mammalian cells. It utilizes electroporation to release cytosolic proteins, including lactate dehydrogenase (LDH). The sequestered LDH and total cellular LDH fractions are determined to assess LDH sequestration, providing insights into the overall bulk sequestration activity.

An Assay for the Quantification of Inorganic Polyphosphate in Bacteria

Source: Pokhrel, A., et al. Assaying for Inorganic Polyphosphate in Bacteria. J. Vis. Exp. (2019).This video demonstrates a colorimetric assay to measure inorganic polyphosphate (polyP) in bacterial samples. The procedure involves enzymatically converting polyP into free phosphate, which then reacts with molybdate and ascorbic acid to produce a blue-colored complex. By comparing the absorbance of this complex to a standard curve of known phosphate concentrations, the polyP content in bacterial...
