Humanized Mouse

A humanized mouse is an immunodeficient mouse engrafted with human cells, tissues, or immune components to model human biology in vivo. In cancer research, transplanted human hematopoietic stem cells can develop into human immune-cell populations, while patient-derived tumors or cancer cells establish disease models within the host. These models help researchers study tumor–immune interactions, evaluate immunotherapies, and assess how human cancers respond to drugs in a living system. By linking human cellular biology with controlled animal experiments, humanized mice support preclinical research and may improve the relevance of therapeutic testing before clinical studies.

Here, we present a novel humanized mouse liver model generated in Alb-toxin receptor mediated cell knockout (TRECK)/SCID mice following the transplantation of immature and expandable human hepatic stem cells.

Developing a Humanized Mouse Model with Human Liver Cells and Immune Cells

2025

This video demonstrates a method for generating a dual humanized mouse model with a human immune system and liver cells. It involves transplanting human liver cells and hematopoietic stem and progenitor cells into a transgenic immunodeficient mouse via intrasplenic injection, leading to engraftment of liver cells and development of human immune cells.

A Technique to Generate a Human PBMC-Engrafted Humanized Xenograft Mouse Model

2025

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

A Technique for Human Skin Grafting in a Mouse Model

2025

This video demonstrates a method for grafting human skin onto an immunodeficient mouse with a suppressed neutrophil response. The method involves applying human split-thickness skin to the mouse's graft bed, sealing it, and covering it for recovery. Eventually, the human skin adheres to the mouse's skin, establishing vascularization and circulation, resulting in a successful skin xenograft.

Constructing a Humanized Mouse Model with Engineered Immunity against HIV

2025

The video demonstrates a technique to generate a humanized mouse model with stem-cell-based engineered immunity against the human immunodeficiency virus (HIV). Human fetal thymus tissue and engineered hematopoietic stem cells (HSCs) are implanted into the kidney capsule of a severely immunocompromised mouse. The HSCs, which express chimeric antigen receptors (CARs) against HIV, are mixed in a gelatinous protein mixture that promotes their co-localization with thymus tissue. Post-procedure, the...
