Mouse Organoid Media

Mouse organoid media are specialized nutrient and signaling formulations that support the growth and maintenance of three-dimensional mouse organoids, making them essential for modeling tissue biology in the laboratory. Their components provide nutrients, growth factors, hormones, and pathway modulators that regulate cell survival, stem or progenitor cell proliferation, and differentiation within an extracellular matrix environment. In cancer research, optimized media help establish organoids from healthy or tumor tissue, preserve key cellular features, and create reproducible models for studying tumor development, drug responses, and treatment resistance. In vitro Organoid Culture of Primary Mouse Colon Tumors

A simple method to establish primary murine colon tumor organoid is described. This method utilizes the feature that colon tumor cells survive and grow into organoids in media containing limited growth factors, whereas normal colon epithelial do not.

This protocol describes the steps for cloning multiple single guide RNAs into one guide RNA concatemer vector, which is of particular use in creating multi-gene knockouts using CRISPR/Cas9 technology.
