Subcutaneous Tissue

Subcutaneous tissue is the layer of connective and adipose tissue beneath the dermis that anchors the skin to underlying structures, cushions the body, and helps regulate heat and energy storage. Its extracellular matrix, blood vessels, immune cells, and fat cells create a dynamic tissue environment that supports signaling, nutrient exchange, and tissue repair. In cancer research, this compartment is studied as a potential site of tumor growth, local invasion, inflammation, and metastatic spread, as well as a route for administering injectable therapies. Understanding its cellular interactions can clarify how tumors respond to their surrounding microenvironment and improve models of cancer progression and treatment.

We standardized an abdominal adipose tissue biopsy using a non-diathermy method performed under local anesthesia. Three cases of excessive post-operation bleeding out of 115 operations (2.61%) occurred. We conclude that an abdominal adipose tissue surgical biopsy using a non-diathermy method can be safely applied to healthy men.

This protocol describes the isolation of pig adipose-derived stem cells (pADSC) from subcutaneous adipose tissues with examination of multipotency. The multipotent pADSC are used to delineate processes of adipocyte differentiation and study transdifferentiation into multiple cell lineages of mesodermal mesenchyme or further lineages of ectoderm and endoderm for regenerative studies.

The manuscript and associated video demonstrate a percutaneous biopsy technique to obtain samples of subcutaneous adipose tissue from areas surrounding the umbilicus. This method is a low-risk and efficient way to investigate a range of parameters (e.g., gene or protein expression, enzyme activity, lipid content) within adipose tissue.

Inguinal Subcutaneous White Adipose Tissue (ISWAT) Transplantation Model of Murine Islets

In this protocol, a method of murine islet isolation and transplantation into the inguinal subcutaneous white adipose tissue is described. Isolated syngeneic murine islets are transplanted into a murine recipient using a basement membrane hydrogel. The blood glucose level of the recipients is monitored, and histology analysis of the islet grafts is performed.

Subcutaneous Trigeminal Nerve Field Stimulation for Refractory Facial Pain

Treating chronic or neuropathic facial pain can be challenging when medical or standard treatment fails. Subcutaneous nerve field stimulation is the least invasive form of neuromodulation and is used for chronic back pain. We applied this technology to treat chronic and neuropathic trigeminal facial pain.
