Zebrafish Larvae Injection

Zebrafish larvae injection is a microsurgical technique that delivers cancer cells, drugs, or other materials into precise anatomical sites of developing zebrafish to model tumor biology in vivo. Using a fine glass needle and micromanipulator, researchers introduce material into locations such as the yolk sac, circulation, or surrounding tissues, where fluorescently labeled cells can be tracked during development. In cancer research, this approach supports rapid assessment of tumor growth, invasion, dissemination, angiogenesis, and treatment response in a transparent, genetically tractable organism. Its small size, accessible embryonic stages, and compatibility with imaging and drug screening make it valuable for investigating cancer mechanisms and evaluating candidate therapies.

Inducing Neuroinflammation in Zebrafish Larvae with a Lipopolysaccharide Injection

2025

The video demonstrates the microinjection of lipopolysaccharides (LPS) into the brains of zebrafish larvae to induce neuroinflammation. The larvae are positioned on an agarose-lined dish, the needle is inserted into the brain ventricles, and the LPS is injected. The injected LPS triggers an immune response that leads to neuroinflammation.

We present a novel method for microgavage of larval zebrafish utilizing standard embryo microinjection and stereomicroscopy equipment. We demonstrate that microgavage is a safe and efficient technique useful for delivering controlled amounts of diverse materials specifically into the larval zebrafish intestinal lumen.

Here, we present a detailed protocol for automated tumor xenograft implantation into zebrafish larvae using a state-of-the-art robotic injection system. Furthermore, we present injection videos and validation data demonstrating the system's successful performance.

A Technique to Quantify Fungal Infection in Zebrafish Larvae

2025

This video demonstrates a method to quantify fungal infection in zebrafish larvae. Fungal spores are microinjected into the larval hindbrain ventricle, and the larvae are incubated to allow infection development. Infected larvae are harvested at defined intervals, and infection progress is assessed by enumerating colony-forming units.

The rapid development, small size and transparency of zebrafish are tremendous advantages for the study of innate immune control of infection1-4. Here we demonstrate techniques for infecting zebrafish larvae using the fungal pathogen Candida albicans by microinjection, methodology recently used to implicate phagocyte NADPH oxidase activity in control of fungal dimorphism5.
