Alpha Substitution

Alpha substitution is an organic chemistry reaction in which a hydrogen or other substituent on the carbon adjacent to a carbonyl group is replaced by a new group. The process commonly begins when a base removes an acidic alpha hydrogen, forming a resonance-stabilized enolate whose carbon reacts with an electrophile; protonation or other workup then gives the substituted carbonyl compound. Depending on the electrophile and reaction conditions, alpha substitution can introduce alkyl, acyl, or halogen groups. These reactions provide a practical way to construct carbon-carbon bonds and increase molecular complexity in synthesis, including the preparation of pharmaceuticals, natural products, and other functional organic molecules.
