Isotope Mass Analysis

Isotope mass analysis is the measurement of the masses and relative abundances of an element’s isotopes, providing chemical information beyond average atomic mass. In mass spectrometry, a sample is ionized, and the resulting ions are separated according to their mass-to-charge ratios before detection; the isotope pattern or ratio is then interpreted to identify substances and determine composition. In chemistry, this approach supports molecular characterization, elemental analysis, reaction tracing, and the study of natural or artificially enriched isotope distributions. Its quantitative measurements also help investigate reaction pathways, sample origins, and changes in chemical systems.

This protocol will demonstrate the extraction and analysis of free and esterified bioactive fatty acids from cells. Fatty acids are accurately quantified using stable isotope dilution, chiral liquid chromatography, electron capture atmospheric chemical ionization multiple reaction monitoring mass spectrometry (SID-LC-ECAPCI-MRM/MS).

Previous work suggests that the nitrogen isotopic composition of atmospheric nitrogen oxides might distinguish the influence of different sources in the environment. We report on an automated, mobile, field-based method for the high collection efficiency of atmospheric NOx for N isotopic analysis at an hourly time resolution.

Analyses of the sulfur isotopic composition (δ34S) of pyrite from methane-bearing sediments have typically focused on bulk samples. Here, we applied secondary ion mass spectroscopy to analyze the δ34S values of various pyrite generations to understand the diagenetic history of pyritization.
