Monocyte Culture

Monocyte culture is the controlled maintenance of monocytes outside the body to study their survival, differentiation, and immune functions. In culture, isolated monocytes respond to defined media and signaling conditions, including cytokines and microbial stimuli, that can promote differentiation into macrophage-like or dendritic cell-like cells and alter inflammatory mediator production. This approach enables researchers to examine innate immune recognition, pathogen interactions, antigen presentation, and regulation of inflammation in immunology and infection studies. Monocyte cultures also provide an accessible model for comparing donor responses, testing immune-modulating compounds, and investigating how cellular environments influence host defense and disease-related immune activity.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.

Isolation of Human Monocytes by Double Gradient Centrifugation and Their Differentiation to Macrophages in Teflon-coated Cell Culture Bags

We present a simple and efficient protocol for the generation of human macrophages. Buffy coats are processed by double density gradient centrifugation and isolated monocytes are then differentiated to macrophages in Teflon-coated cell culture bags. This maximizes macrophage yields and facilitates cell harvesting for subsequent experiments.
