Mycobacteria Enumeration

Mycobacteria enumeration is the quantitative measurement of mycobacterial cells in a sample, providing essential information about bacterial burden, growth, and infection severity. Viable organisms are commonly estimated by dispersing cell clumps, preparing serial dilutions, and plating samples for incubation before counting colony-forming units; staining or molecular assays can measure total mycobacterial material. Because the lipid-rich cell envelope promotes aggregation and mycobacteria often grow slowly, sample preparation, incubation conditions, and counting criteria strongly influence accuracy. In immunology and infection research, enumeration supports evaluation of host control, antimicrobial activity, disease progression, and experimental infection models.

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.
