Threading Intercalator Inhibition

Threading intercalator inhibition is a molecular strategy in which a compound inserts an aromatic core between adjacent DNA base pairs while its attached groups remain positioned along the nucleic acid backbone. Unlike simple intercalators, threading molecules pass through a transiently opened base-pair step, creating a stable DNA complex that can hinder polymerase movement, replication, or transcription. In immunology and infection research, this mechanism helps explain how DNA-targeting compounds suppress microbial proliferation and provides a framework for evaluating antimicrobial activity, selectivity, and toxicity. Studying these interactions also supports the design of inhibitors that interfere with pathogen genome maintenance.
