Treg Isolation

Treg isolation is the process of separating regulatory T cells from mixed immune-cell populations for functional and molecular study. In immunology, researchers typically enrich CD4+CD25+ cells using antibody-coated magnetic beads, then improve purity by flow-cytometric sorting with markers such as CD4, CD25, and FOXP3 while limiting contamination by conventional T cells. Isolated Tregs can be assessed in suppression assays to determine how effectively they inhibit responder T-cell proliferation and cytokine production. This approach supports research into immune tolerance, infection, autoimmunity, transplantation, and cancer, where altered Treg abundance or activity can influence disease progression and therapeutic responses.

With effector functions distinct from other T cell subsets, Th17 cells have been centrally implicated in inflammatory autoimmunity. This in vitro Th17 differentiation protocol provides a means to determine whether naïve CD4+ T lymphocytes can differentiate into Th17 cells, and to further examine their role in autoimmunity and host response.

Naïve CD4+ T cells polarize to various subsets depending on the environment at the time of activation. The differentiation of naïve CD4+ T cells to various effector subsets can be achieved in vitro through the addition of T cell receptor stimuli and specific cytokine signals.

We describe the isolation of cardiac extracellular matrix from C57Bl/6J control mice, tight-skin mice, and tight-skin mice treated with the IRF5 inhibitory peptide. We also describe the vasodilation studies on the isolated vessels from C57Bl/6J, tight-skin mice and tight-skin mice treated with the IRF5 inhibitory peptide.

Neutrophils are among the first cells to arrive on the site of inflammatory immune response, and their functions and mechanisms have been studied extensively in vitro. We demonstrate a standard density gradient separation method to isolate human neutrophils from whole blood using commercially available separation media.
