Cecum Puncture

Cecum puncture is a medical and experimental procedure in which a needle or other instrument penetrates the cecum, the first section of the large intestine, to access its contents or create a controlled injury. In the cecal ligation and puncture model, puncturing the ligated cecum allows fecal material and intestinal bacteria to enter the peritoneal cavity, producing polymicrobial peritonitis, systemic inflammation, and potentially sepsis. Researchers use this approach primarily in preclinical medicine to study host responses to infection, evaluate antibiotics and supportive therapies, and investigate organ dysfunction associated with sepsis. Its controlled design supports comparisons among disease mechanisms and treatment strategies.

Experimental sepsis can be induced in mice using the cecal ligation and puncture (CLP) method. Current protocols to assess autophagy in vivo in the context of CLP-induced sepsis are presented here: A protocol for measuring autophagy using (GFP)-LC3 mice, and a protocol for measuring autophagosome formation by electron microscopy.
