Mirna Network

A miRNA network is a regulatory system in which microRNAs, small noncoding RNAs, coordinate the expression of many genes and influence cellular functions relevant to health and disease. After processing into mature miRNAs, they associate with Argonaute-containing RNA-induced silencing complexes and recognize complementary sequences, often in messenger RNA 3′ untranslated regions, to reduce translation or promote transcript degradation. Because individual miRNAs can regulate multiple targets and several miRNAs can control one transcript, these networks integrate signaling pathways, feedback loops, and cell-state responses. In medicine, miRNA networks support biomarker development, disease mechanism studies, therapeutic target identification, and investigations of cancer, cardiovascular disorders, and other conditions.

miRNA Extraction: A Method to Extract miRNA from Plasma Sample

2023

MicroRNAs (miRNAs) are tissue specific, small, non-coding RNAs regulating gene expression, and could represent ideal candidates for early detection of lung cancer. In the featured protocol, we will isolate microRNA from patient blood samples for early lung cancer screening.

Circulating miRNA Extraction: A Method to Isolate miRNA from Plasma Samples Via Organic Extraction and Small RNA Enrichment

2023

This video describes the extraction of circulating microRNA from plasma samples of colorectal cancer patients using organic extraction followed by glass-fiber column enrichment. miRNAs are stable in biological fluids such as serum and plasma, which renders them ideal circulating biomarkers for cancer diagnosis, prognosis, and treatment decision-making and monitoring. This method will help identify new circulating biomarkers capable of predicting clinical outcomes in patients.

Modelling Intratumoral Treatment with miRNA Inhibitors: A Method to Analyze the Effect of miRNA Inhibitors on Tumor Growth in Murine Model

2023

This video demonstrates the in vivo delivery of miRNA inhibitors as therapeutic agents against tumor growth in orthotopic mouse models. The intratumoral administration of synthetic miRNA inhibitors or antagomiRs allows them to bind and inhibit endogenous miRNAs, thereby causing mRNA upregulation and suppression of tumor growth.

2023

miRNA or microRNA are key regulators of cell cycle progression. This video describes a flow cytometry-based cell cycle analysis of lung cancer cells transfected with miRNA, to study the effect of miRNA on cell cycle regulation. Inhibition of proliferation by halting cell cycle progression has shown potential to be a promising approach for cancer treatment.
