Miniature Endplate Potentials

Miniature endplate potentials (MEPPs) are small, spontaneous depolarizations of a muscle fiber’s membrane at the neuromuscular junction, providing a measurable signal of quantal neurotransmitter release. They occur when a single synaptic vesicle releases acetylcholine into the synaptic cleft, activating nearby nicotinic acetylcholine receptors and producing a brief endplate response, even without an action potential in the motor neuron. Recording MEPP frequency and amplitude helps distinguish changes in presynaptic vesicle release from postsynaptic receptor function. In neuroscience, these measurements support the study of synaptic transmission, neuromuscular development, plasticity, and disorders affecting communication between motor neurons and muscle.

A reverse engineering system is employed to record and obtain detailed and comprehensive geometry data of vertebral endplates. Parametric models of vertebral endplate are then developed, which are beneficial to designing personalized spinal implants, making clinical diagnoses, and developing accurate finite element models.

The video demonstrates the electrophysiological analysis of embryonic stem cell-derived neurons after exposure to neurotoxins. Treating these neurons with botulinum neurotoxins reduces the frequency of spontaneous miniature excitatory post-synaptic currents, confirming inhibition of synaptic transmission.

Here we describe electrophysiological methods for measuring synaptic transmission at the neuromuscular junction of Drosophila larva. Evoked release is initiated artificially by stimulating the motor neuron axons, and transmission through the NMJ can be measured by the postsynaptic response evoked in the muscle.
