Murine Cervical Heart Transplantation Model Using a Modified Cuff Technique

The murine cervical heart transplantation model is well suited for immunological as well as ischemia reperfusion injury studies. We modified the procedure using a non-suture cuff technique and performed more than 1,000 successful transplants with this approach.
Herein, we provide additional details of this technique to supplement the video.
I am UBA from the Daniel KY Research Laboratory of the Department of Visceral Transplant and Thoracic Surgery of the Medical University of Ensberg. Today we are going to show you a Morine heterotopic heart transplant model. Using a Modified cuff technique To recipient animal is anesthetized with an intramuscular injection of INE and ketamine.
Next, all hair of the cervical lateral region are removed and the operative field scrapped three times with chlorhexidine. Then the surgical field is prepared and the animal is placed in a supine position on the operative field, the donor mouse is anesthetized with an intramuscular injection oxil sine and ketamine. Next, all hair of the abdominal region of the recipients are removed and the operative field scrapped three times with chlorhexidine.
Next, the surgical field is prepared and the animal is placed in a supine position on the operative field. As shown in this sequence, cardiac Carlo grafts from biopsy donors are rejected within seven days after transplantation into fully MHC mismatched C 57 black six recipients. Syngeneic heart transplants performed in our series survived indefinitely.
Despite the fact that the heterotopic heart transplantation in mice is technically challenging, it remains the gold standard for any mechanistic in vivo studies related to transplant immunology. Our modified approach allows achieving high success rates with basic microsurgical skills that will allow broad application of this model. This video aims to help and guide other researchers to establish the model in their own laboratories.
