Perspectives on Neuroscience

As usual, the main cause for progress in natural sciences are methodological developments. Of course, conceptual developments follow and go in parallel, but I think the, the tremendous expansion of the field of neurosciences was triggered by a number of methodological breakthroughs at the lowest level. These were the progresses in the domain of molecular biology, genetic engineering, which provided model systems for the study of development for the study of certain diseases. These techniques also provided extremely valuable tools for the biochemical and molecular identification of neurons. We learned about a degree of diversification of morphologically seemingly identical neurons that are endowed with different molecular machineries. All this we owe to the development of molecular biology.A second important factor I think was the development of non-invasive imaging technologies. cause this allowed us to transfer a large number of concepts that have been developed in animal experimentation to investigations in human subjects. So this is the functional magnetic resonance imaging pository emission tomography. Also the introduction of the magneto ence holography, which allowed for much b

We introduce a novel workflow for electron microscopy investigations of brain tissue. The method allows the user to examine neuronal features in an unbiased fashion. For elemental analysis, we also present a script that automatizes most of the workflow for randomized sampling.

An Unbiased Approach of Sampling TEM Sections in Neuroscience

The protocol for conducting fNIRS hyperscanning experiments on collaborative learning dyads in a naturalistic learning environment is outlined. Further, a pipeline to analyze the Inter-Brain Synchrony (IBS) of oxygenated hemoglobin (Oxy-Hb) signals is presented.

Inter-Brain Synchrony in Open-Ended Collaborative Learning: An fNIRS-Hyperscanning Study

This study presents a naturalistic experimental setup that allows researchers to present real-time action stimuli, obtain response time and mouse tracking data while participants respond after each stimulus display, and change actors between experimental conditions with a unique system including a special transparent organic light-emitting diode (OLED) screen and light manipulation.

A Naturalistic Setup for Presenting Real People and Live Actions in Experimental Psychology and Cognitive Neuroscience Studies

This protocol demonstrates how physics-based neural simulations can be used to interpret electrophysiological biomarkers of neurotherapeutics and uncover their effect on neural circuits, providing a mechanistically grounded approach for neurotherapeutic development.

A Protocol For Uncovering Neural Mechanisms Of Neurotherapeutic Effects On Electroencephalography Using The Human Neocortical Neurosolver

The ability to culture cells has revolutionized hypothesis testing in basic cell and molecular biology research. It has become a standard methodology in ...

BioMEMS and Cellular Biology: Perspectives and Applications

Genetic studies in yeast can be employed to investigate the molecular and cellular functions of human genes in cellular DNA metabolism. Methods are described for the genetic characterization of the human WRN gene product defective in the premature aging disorder Werner syndrome in functionally conserved pathways using yeast as a tractable model system.

Genetic Studies of Human DNA Repair Proteins Using Yeast as a Model System

This article details the procedures involved in overexpression and analysis of small GTPases in polarized epithelial cells using microinjection technique.

The cytochrome c oxidase/sodium dehydrogenase (COX/SDH) double-labeling method allows for direct visualization of mitochondrial respiratory enzyme deficiencies in fresh-frozen tissue sections. This is a straightforward histochemical technique and is useful in investigating mitochondrial diseases, aging, and aging-related disorders.

Visualization of Mitochondrial Respiratory Function using Cytochrome C Oxidase / Succinate Dehydrogenase (COX/SDH) Double-labeling Histochemistry

Single cell expression profiling allows the detailed gene expression analysis of individual cells. We describe methods for the isolation of cardiomyocytes, and preparing the resulting lysates for either whole transcriptome microarray or qPCR of specific targets.

Single Cell Transcriptional Profiling of Adult Mouse Cardiomyocytes

Glycolysis is a defining metabolic marker in multiple biological systems. Monitoring glycolysis by measuring the extracellular flux of H+ is common, but requires correction to be quantitative and unambiguous. Here, we demonstrate how to gather and correct extracellular flux data to distinguish between respiratory and glycolytic sources of extracellular acidification.
