Summary
Trace eyeblink klassieke conditionering (ECC) werd gebruikt om te beoordelen hippocampal-afhankelijke associative leren bij volwassen ratten die waren toegediend een hoge concentratie (11,9% v/v) van alcohol tijdens de vroege ontwikkeling van de neonatale hersenen. In het algemeen, zijn ECC procedures diagnostische hulpmiddelen voor het opsporen van hersenen dysfunctie over vele psychologische en biomedische instellingen voor geluid.
Abstract
Neonatale ratten werden toegediend een relatief hoge concentratie van ethylalcohol (11,9% v/v) tijdens postnatale dagen 4-9, een tijd waarin de foetale hersenen snelle organisatorische veranderingen ondergaat en is vergelijkbaar met versnelde hersenen veranderingen die zich tijdens het derde trimester bij de mens voordoen. Dit model van fetal alcohol spectrum disorders (FASDs) produceert ernstige schade aan de hersenen, het nabootsen van het bedrag en de patroon van binge-drinken dat zich in sommige zwangere alcoholische moeders voordoet. We beschrijven het gebruik van trace eyeblink klassieke conditionering (ECC), een variant van de hogere-orde van associative leren, om te beoordelen op lange termijn hippocampal dysfunctie, die meestal in volwassen nakomelingen alcohol-blootgesteld wordt gezien. Op de leeftijd van 90 dagen, knaagdieren waren operatief bereid met opname en stimulerende elektroden, die gemeten Electromyografische (EMG) knipperen van de activiteit van de spier links ooglid en milde schok posterieure voor het linker oog, respectievelijk afgeleverd. Na een herstelperiode van 5 dagen onderging ze 6 sessies van trace ECC associative leren verschillen tussen alcohol-blootgesteld en controle van de ratten te bepalen. Trace ECC is één van vele mogelijke ECC procedures die moeiteloos kunnen worden gewijzigd met behulp van dezelfde apparatuur en software, zodat verschillende neurale systemen kunnen worden beoordeeld. ECC procedures in het algemeen, kunnen dienen als diagnostische hulpprogramma's voor het opsporen van neurale pathologie in de hersenen van de verschillende systemen en verschillende omstandigheden die beledigen van de hersenen.
Representative Results
De eyeblink-software is geschikt voor een grote en uitgebreide verzameling van gegevens te voorzien van vele soorten metingen. Voor beknoptheid, rapporteren we in deze studie, vertegenwoordiger resultaten voor leren en prestaties maatregelen waaronder adaptieve CR percentage, adaptieve CR amplitude, UR percentage en UR amplitude. De adaptieve CR-periode werd gekozen als het vertegenwoordigt de verwerving van een goed getimede eyeblink reacties over herhaalde opleiding, als gevolg van verbeterde synaptische plasticiteit in de hippocampus tijdens trace ECC50,51,52. De UR-maatregelen werden gekozen om te verhelderen of de neonatale alcohol-geïnduceerde leren tekorten in ECC waren als gevolg van storingen in associative leren of verstoringen traceren in het reageren op de schok ons - die op motiverende of motor verschillen, in plaats van leren verschillen tussen behandelgroepen wijzen kunnen. Gegevens voor elke maatregel zijn geanalyseerd met behulp van 2 (seks) x 3 (neonataal groep) x 6 (sessie) gemengd ANOVAs, met een sessie als de factor herhaald-maatregelen. Belangrijke belangrijkste gevolgen voor neonatale behandeling zijn geanalyseerd met behulp van Tukey post hoc tests en belangrijke interacties werden geanalyseerd met behulp van eenvoudige effecten proeven. Alle statistische analyses werden uitgevoerd met een minimale alfaniveau van 0,05 en resultaten in grafieken zijn gemiddelde ± SEM.
Beginnend met de adaptieve CR percentage maatregel, de ANOVA aangegeven een belangrijke belangrijkste effect van neonatale groep, F(2,21) 11.69, p = < 0.001, maar geen significant belangrijkste effect van geslacht (p = 0.71) of belangrijke interactie tussen deze elementen (p = 0,20). Zoals verwacht, adaptieve CR percentage verhoogd de zes sessies van opleiding, F(5, 105) = 81.15, p < 0.001, en de verschillen tussen de neonatale groepen waren afhankelijk van enige mate van sessie, F(10, 105) = 4,58, p < 0,001. Er waren geen andere belangrijke interacties met betrekking tot de factor van de sessie. Ook voor adaptieve CR amplitude, was er weer een aanzienlijke belangrijkste effect van neonatale groep, F(2,21) 22.32, p = < 0.001, maar geen significant belangrijkste effect van geslacht (p = 0.21) of belangrijke interactie tussen deze elementen (p = 0,48). CR amplitude ook aanzienlijk toegenomen de zes sessies van opleiding, F(5, 105) = 59.27, p < 0.001, en de verschillen tussen de neonatale groepen waren afhankelijk van enige mate van sessie, F(10, 105) 4.31, p = < 0,001. Globaal, beide maatregelen CR toonde significante verschillen tussen de groep middelen en deze middelen gescheiden aanzienlijk bij de verschillende sessies van de opleiding. Om te bevestigen welke groepen verschilde aanzienlijk, van Tukey post hoc tests bleek dat de ratten (AI) alcohol-intubated uitgevoerd aanzienlijk slechter op beide CR maatregelen dan de unintubated-control (UC) en de sham-intubated (SI) ratten (p < 0,01 voor CR percentage; p < 0.001 voor CR amplitude), die niet van elkaar verschillen (p's> 0,05). Eenvoudige effecten proeven uitgevoerd op de aanzienlijke neonataal groep x sessie interacties voor beide maatregelen CR, bevestigd dat de AI rats belangrijker is in het verwerven van CRs begint bij sessie 2 en t/m 6 van de sessie op te voeren ten opzichte van zowel UC en SI ratten werden aangetast (alle p's< 0,05), die niet van elkaar verschillen gedurende zes sessies. De enige uitzondering was adaptieve CR amplitude SI ratten begonnen niet significant afwijken van AI ratten tot sessie 3. Deze resultaten worden weergegeven in figuur 5A, 5B.
Er waren geen significante verschillen in de UR maatregelen als gevolg van seks, neonatale groep of interacties van deze factoren met de factor van de sessie. Deze negatieve bevindingen aangegeven dat elke groep was in staat om het uitzenden van eyeblink reacties op de schokken ons even, en dat de tekorten van de leren waargenomen in de AI-ratten niet werden beïnvloed door motiverende of motor verschillen in knipperen (figuur 6A, 6B).

Discussion
Neonatale rat pups die ethylalcohol tijdens postnatale dagen 4-9 ontvangen tentoongesteld trace eyeblink conditioning bijzondere waardeverminderingen in de volwassenheid. Deze bevindingen ondersteunen de idee dat alcohol een teratogen is met blijvende schadelijke gevolgen voor de hippocampal functie. Algemene, geconditioneerde reageert in de trace-procedure was lager voor ratten blootgesteld aan alcohol in vergelijking met ratten in beide controlegroepen. De waardeverminderingen op associative leren in de rats alcohol-blootgesteld werden niet beïnvloed door motiverende of motor verschillen (dwz., geen verschillen in het knipperen aan schok U.S. intensiteit).
Terwijl trace ECC een handig diagnostisch hulpmiddel voor informatief uitdaging-geïnduceerde hippocampal Neuropathologie is, moeten de resultaten van deze methode worden geplaatst in de juiste context. Ten eerste, de belangrijkste procedurele elementen in deze demonstratie betrokken de gerichte levering van alcohol tijdens een bekende venster van de kwetsbaarheid voor de ontwikkelende hersenen, fabricage van elektrode hardware waarmee opname van Electromyografische activiteit en levert shock, chirurgische implantatie van de bovengenoemde hardware en latere dierproeven met behulp van een leren paradigma dat een cognitieve functie van belang beoordeelt. In elke fase in het proces, moet worden gezorgd om geen onnodige/onbedoelde schade toebrengen aan de knaagdier onderwerpen en regelmatig te controleren hun gezondheidssignalering. Hun gedrags resultaten bieden de "venster" naar cognitie, een psychologische construct dat alleen nauwkeurig beschreven wanneer hun gezondheid niet in gevaar komt door experimentele fouten omvat alcohol doseren, hardware defecten of chirurgische implantatie. Elk procedurele element in het onderzoeksproces moet dus worden uitgevoerd op een verantwoorde wijze om ervoor te zorgen dat de resultaten van ECC kunnen worden geëxtrapoleerd naar de bevindingen bij de mens. Ten tweede, het ECC paradigma geeft inzicht over de aard van de associative leren, maar wees voorzichtig niet om uit te breiden bevindingen met behulp van deze aanpak in grote lijnen het toeschrijven van hen aan andere cognitieve domeinen - zoals werkgeheugen, korte/lange termijn herinneringen en bewustzijn - tenzij een sommige facet van deze domeinen binnen een ECC studie door experimenteel design heeft opgenomen. Bijvoorbeeld, deze demonstratie onderzocht de fase overname van trace ECC leren, maar heeft niet onderzocht geheugen bewaren in de rats nadat zij de opleiding voltooid. Geheugen is dus een onafhankelijke psychologische proces dat naast leren moet worden geëvalueerd. Bij het ontwerp, kan een een geheugen retentie interval om te beoordelen of vermogen op korte termijn of lange termijn geheugen opnemen. Ten derde is zijn er parallelle geheugen systemen54 die gelijktijdig kunnen werken samen met motiverende, ervaringsgerichte en hormonale factoren die aan gedrag bijdragen, erkenning essentieel voor dien verstande dat associativiteit (tijdens ECC) is maar een van de vele processen die wat is onthullen "goed" of "slecht" over leren. Tot slot, trace ECC is niet een zuiver hippocampal-afhankelijke taak, zoals andere hersengebieden sommige component van de CR kunnen bemiddelen. Een goed begrip van de interacties tussen de verschillende neurale circuits en/of het soort stimulans parameters die worden gebruikt in een studie, moet dus rekening worden gehouden bij het maken van de gevolgen die berust op discrete resultaten. Het cerebellum, bijvoorbeeld, draagt ook bij aan trace ECC, waar het beïnvloedt de topografische kenmerken van de CR en CR timing, met name wanneer de ISI is kort van duur. Trace ECC niet wordt beïnvloed bij mensen met cerebellaire schade die worden getest met een lange trace interval (1000 ms), maar bij degenen die een kortere trace interval (400 ms)34ontvangen wordt beïnvloed. Bovendien voorkomen bilaterale laesies van de dorsale mediale prefrontale cortex (mPFC) die gericht zijn op de regio anterior cingularis en mediale agranular's in muizen, overname van trace CRs55, terwijl de vernietiging van de caudal mPFC bij konijnen produceert soortgelijke resultaten46. Deze bevindingen benadrukken ook het belang rekening te houden met soorten verschillen in de prefrontale bijdragen aan cerebellaire-hersenen het gevolg gedreven associative leren, zoals trace ECC. Terwijl neonatale alcohol blootstelling tijdens PD 4-9 aangetast verwerving van 500-ms trace CRs voor volwassen in deze studie en anderen47,56 ratten, is dit niet het zelfde geval voor neonatale alcohol-blootgesteld ratten die zich voordoen van een 300-ms trace interval, zelfs wanneer uitgedaagd bij een relatief hoge dosis van alcohol (5 g/kg)57, suggereert dat de waardevermindering van de trace bij alcohol-blootgesteld ratten afhankelijk van de duur van het interval trace is.
In deze studie werd de hippocampus als uitermate belangrijk voor het bemiddelen van trace ECC, en als uitgedaagd door neonatale alcohol blootstelling, neurale-gerelateerde schade vertoont, zoals weerspiegeld door waardeverminderingen in overname van trace CRs benadrukt. Het moet waarschuwde echter dat het cerebellum-hersenen stam circuits, met name de interpositus kern, is essentieel voor de vele facetten van ECC, waaronder de verwerving, expressie en topografische kenmerken van de CR, afhankelijk van het type van taak van de ECC met inbegrip van trace ECC36,40,55,58,59. Inderdaad, dit neurale circuit interageert met de hippocampus voor het besturen van de expressie van CRs tijdens hogere-orde vormen van ECC, zoals trace ECC60. Het is niet geheel duidelijk of alcohol blootstelling tijdens de vroege ontwikkeling van de hersenen specifiek hippocampal functie in trace ECC beïnvloedt. Veel verschillende hersengebieden zijn kwetsbaar voor vroege alcohol belediging, met inbegrip van de mPFC, het cerebellum en de hippocampus18,19,23,47,61,62, en het is zeer waarschijnlijk dat alcohol verstoort de werking van deze structuren voor wisselend en variërend, maar functioneel belangrijke verschillen over vele ECC procedures. Ondanks de valkuilen betreffende de interpretatie van de resultaten van de trace ECC studies, is succesvolle overname van trace CRs aangetoond ten minste vertrouwen op een intact hippocampus, zoals ondersteund door dierlijke laesie studies42,44,63,64,,65. Deze procedure blijft dus een zeer waardevolle benadering voor het aantonen van de verbanden tussen ontwikkelings alcohol blootstelling aan trace geconditioneerd reageren omdat de neurale circuits die ten grondslag liggen aan het, is veel beter begrepen dan die van andere hippocampal-afhankelijke taken, zoals het leren van de plaats in de Morris water maze, roman objectherkenning, en contextuele- and -trace angst conditionering.
ECC als een gedrags methode om te "assay" cognitie, heeft brede toepasbaarheid op het gebied van developmental neuroteratology. Inderdaad, recente bevindingen uit ons lab ondersteunen het idee dat de ontwikkelingslanden hippocampus is zeer gevoelig voor de effecten van alcohol, die kunnen worden verzacht door verschillende Interventionele strategieën18,47. Het belangrijkste voordeel hier is dat met een beter begrip van de alcohol-geïnduceerde trace ECC leren tekorten, kunnen zij voorspellende van andere problemen in de hippocampal gebaseerde functies buiten associative leren - met name die bekend worden gemedieerd door de zelfde hippocampal neurocircuitry.
Toepassing van trace ECC en andere varianten (bijvoorbeeldvertraging, omkering, discriminatie, samengestelde) ophelderen van de neurobiologische mechanismen en neurale systemen die betrokken zijn bij het leren van de associative, kan worden uitgebreid buiten het gebied van foetaal alcohol onderzoek. Dit paradigma heeft bijvoorbeeld veel aandacht gekregen in de gevallen bij de mens en dierlijke modellen van psychiatrische aandoeningen zoals schizofrenie66,67, neurodegeneratieve ziekten zoals de ziekte van Alzheimer68,69en drugs misbruik70,71,72. De voordelen ervan als een onderzoeksmethode te beoordelen Neurocognitieve functie en dysfunctie zijn dus duidelijk in veel psychologische en biomedische disciplines, met inbegrip van de neurowetenschappen.
